Research Abstract

正常肝細胞が肝癌細胞に変化する過程ではどのような遺伝子変化が起るのかについてはほとんど解っていない。本研究ではこのような現状にふまえて、研究対象をマウス肝癌に絞って遺伝子変化を解析し、その知見をヒト肝発癌のメカニズムの解析に役立てることを目標としている。実験的マウス肝発癌は確立された発癌モデルであり、またマウスは遺伝子情報が豊富であること、短期間に多数繁殖するために遺伝学的解析が容易であること、細胞培養が容易であることなどの利点がある。本研究グループではこのような利点を活かしてマウス肝発癌の分子機構を新しい分子遺伝学的手法を用いて様々な角度から解析した。
本年度は、以下の点が明らかになった。
1. RLGS法、MS-RDA法によりマウス肝癌でメチル化の異常を示す多数のDNA断片が検出された。その中には癌抑制遺伝子の可能性のあるmac25を含むものが見られ、肝癌ではこの遺伝子の発現が低下していた。また、上記のDNA断片から未知の遺伝子が多数検出された。
2. MSM及びC3HマウスのRLGSマップ完成した。これを応用してC3HxMSMF1マウスのDEN誘発肝癌でも多数の欠失スポットを検出した。
3. マウス肝癌細胞ではIgf2のloss of imprintingが高頻度に見られた。
4. 肝癌では特累な遺伝子発現が観察され、これらにはIL-1 receptor antagonist(IL-1ra)、ATPase inhibitor等が含まれていた。特にIL-1raの発現は増殖能と相関した。
5. マウス肝癌ではβ-cateninのmutationが高頻度に見られ、この変化は多段階発癌の後期に起ることが明らかになった。
