Research Abstract

大腸菌の分泌蛋白質輸送装置は、膜内在性因子SecY,SecEが形成するチャネルに、SecAがATP依存的に分泌蛋白質を送り込むことにより作動している。装置の活性化に重要なSecGは、膜内配向性の反転と復帰を繰り返していることを明らかにした。また、SecYとSecEを平面膜に再構成し、多数のチャネルのコンダクタンスを解析した結果、いずれも約100pSの均一なチャネルであることを示した。さらにSecAとATP,前駆体蛋白質の添加で順次チャネルは閉じることを明らかにした。
