Research Abstract

エリスロポエチン(EPO)の自然標的細胞である赤芽球系細胞において、エリスロポエチン受容体(EPOR)の下流のJAKファミリーとSTATファミリー遺伝子の発現を解析した。検索対象は、フェニルヒドラジンで誘導された貧血症マウスの脾腫から分離された正常とみなされる赤芽球系細胞と、Friend白血病ウィルス(多血症株)で誘導されたマウス脾腫から分離された赤芽球系細胞を用い、それぞれをTER119抗体を指標に分画した標品について純度が得られた。これらの細胞においてEPOR特異抗体で沈降する蛋白をJAKファミリーの抗体で検索したところ、前者では、EPO依存性にJAK2、STAT5がチロシン燐酸化され、STAT5は核内へ移行し、STAT5A、Bが結合するGAS配列、PRE配列と特異的に結合した。後者では、EPOの有無に拘わらず、JAKI、STAT5が恒常的にチロシン燐酸化されていたが、STAT5は核内移行を示さず、従ってSTAT5の結合するGAS、PRE配列とは結合できなかった。この系にEPOを添加したところSTAT5は瞬時に核移行を示し、EPOとの共働ではじめて増殖シグナルを送ることが初めて示され、ウィルス性白血病発症における、ウィルス遺伝情報とサイトカインシグナルの強調した機能発揮が明らかにされた。今後EPO/EPOR、gp55/EPORを介するSTAT5のチロシン燐酸化状態の差を同蛋白の燐酸化、アミノ酸の同定を含め検討したい。
他方、フレンド白血病細胞系ではEPOR下流のRas機能を打ち消すDN-Rasを導入・発現させると分化が誘導され、MAPK回路がこの現象に関与していることが示され、現在、各種阻害剤を用いてその詳細を検討している。
