Research Abstract |
In this study the expression of gene(s) relating to nutrient ion uptake was investigated using rice mutants deficient in nitrate uptake and resistant to cesium. There was difference in the effects of inhibitors for membrane activity on both nitrate and potassium uptake between NUE5O-1, a low nitrate uptake mutant, and Kinmaze (wild type cultivar) was found. This result suggests that NUE5O-1 is a mutant defecting in ion transport system discriminating nitrate and potassium. Mutants resistant to cesium, a toxic analog for potassium, have been shown to be useful for the research on ion uptake mechanism. Six rice mutants resistant to cesium were isolated from gamma ray-mutagenized M2 population. Preliminary characterization showed that the mutants were divided into several groups. Genetics of cesium resistance has not been determined but gene products specific in the mutant roots were obtained by a simplified differential display method. Furthermore, N02/N03 assay kit (Dojindo, Japan) was shown to be a simple method for determining nitrate and nitrite content of leaves. Using the kit, nitrate metabolism in the leaves of M819, a nitrate reductase-defiecnt mutant, and NUE13-1, another low nitrate uptake mutant, was compared to that of their wild type cultivar.
