Research Abstract

The three-dimensional structure of the intracellular secretory components, especially the Golgi apparatus, was visualized in the mouse Brunner'S gland cells by using multiple stainings of fluorescently labeled lectins and a confocal laser scanning microscope (CLSM). In order to distinguish the Golgi stacks of the cis-side, the intermediate, and the trans-side, three lectins were used each of which recognizes the sugar residue added at the right stacks.
Based on our observations, we propose a new three-dimensional model of the Golgi apparatus : the Golgi apparatus forms a large dome over a nucleus, the lacy structure of the cis-stacks forms its outer boundary, and its inner surface is lined with intermediate stacks which also form a lacy network and is further paved with trans-stackes. Secretory granules formed from the trans-side are released into the internal space of the Golgi apparatus and then transported to the apical region through the holes of the lacy network of the Golgi stacks.
The three-dimensional structure of the Golgi apparatus seems to be the case in other cells which are actively synthesizing mucus. Therefore, we are planning to further investigate the three-dimensional structure of the secretory apparatus by using a CLSM and other new technologies.
