Research Abstract

消化管粘膜におけるTrefoil peptide(pS2 gene)発現に関する研究
本研究の目的は,消化性潰瘍やCrohn病あるいは潰瘍性大腸炎の様な慢性潰瘍性病変の治癒過程におけるpS2などのtrefoil peptides(TP)の発現を分子生物学的に検討し,その治療に貢献しようとするものである.胃潰瘍に関しては,ラット胃に酢酸潰瘍を作成し、胃炎についてはインドメタシンンを用いてモデルを作成しその治癒過程を追跡した.単層培養細胞に我々の考案した方法により欠損部を作成し,その治癒過程を解析した.この際細胞増殖と細胞遊走を分離して検討し、この場合におけるTPの発現を組織学的及び生化学的方法を用いて解析した.またEGF,TGFαなどの増殖因子のTP発現に対する影響を検討した.まずin vivoおよびin vitro でのTP蛋白の発現を免疫組織学的及びWestern blottingを用いて検討した.TPのmRNA発現に関しては,pS2geneのcDNAをRT-PCRを用いて作成しているので,それをプローブとしてNorthern blottingより検討した.TPにはpS2gene,hSPをはじめ多くの種類が存在するため,それぞれの部位でのPCR,RT-PCRを用いた遺伝子学的検討が進行中である.特にヒト生検組織は量的に少ないためPCRを用いた検討を行っている.上記の基礎的検討を基盤として,ヒト生検材料及び手術材料においてTPの発現を検討している.
