ヒト甲状腺腫瘍細胞におけるMAPキナーゼカスケードに関する基礎研究とその臨床応用

Research Abstract

ヒト甲状腺腫瘍では幾つかの癌遺伝子の異常、あるいは成長因子受容体の過剰発現などが報告されている。しかし細胞増殖に関与する情報伝達に関する異常は明らかにされていない。MAPキナーゼカスケードは各種細胞において細胞増殖や分化に重要な役割を果たす。そこでヒト甲状腺腫瘍組織におけるMAPキナーゼ(MAPK),その上流に存在するMAPKキナーゼ(MEK),Raf-1の発現、その活性を正常組織で比較した。甲状腺乳頭癌とろ胞癌組織では大部分でMAPK,MEKの発現量、活性ともに正常部に比して数倍に増加しており、これは免疫組織学的にも証明された。Raf-1活性には差がなかった。一方、良性の腺腫やバセドウ病組織ではMAPK,MEK,Raf-1ともに正常と差がなかった。培養した正常甲状腺細胞にIGF-Iなど種々の成長因子を添加した場合にも増殖に先んじてよMAPKが活性化された。癌組織ではGF-I,IGFBP-2 mRNAの発現増加も観察された。以上よりMAPKカスケードは腫瘍発育に関与する可能性が示唆される。今後の方針としてはMAPK,MEKの遺伝子自体に異常があるか否かを明らかにしたい。
