Research Abstract

覚せい剤や麻薬による薬物依存は大きな社会問題であるが,治療法がほとんど確立されていないのが現状である.治療薬の開発が遅れている理由として,依存形成の鍵となるタンパクが明らかになっていないことが挙げられる.そこで,PCR-select cDNAサブストラクション法を用いて,薬物依存形成の鍵となる新規機能分子の探索を試みた.その結果,覚せい剤メタンフェタミンを連続投与されたマウス脳側坐核から新規機能分子`shati'を同定した.昨年度までに、shatiが,メタンフェタミン誘発自発運動量亢進および場所嗜好性の形成に対して抑制的に働くことを明らかにした.さらに,shatiがメタンフェタミンによって引き起こされる細胞外ドパミン量の増加およびドパミン再取り込み能低下を抑制することを示した.本年度は,メタンフェタミン依存形成段階においてshatiが関与していることを検討するに留まらず,薬物依存抑制因子として報告されているTNF-αとの関連についても明らかにした.PC12細胞へshati発現ベクターをトランスフェクションすると,コントロール細胞と比較して,shatiおよびTNF-αmRNAの発現量の増加,ドパミン再取り込み能の促進,メタンフェタミン誘発ドパミン再取り込み能低下の抑制が観察された.これらshatiの作用は,可溶性TNF受容体およびTNF-α抗体を前処置しTNF-αを中和することよって抑制されたことから,shatiはTNF-αを介してドパミン再取り込みを促進していると考えられる.
今後,shatiの依存形成メカニズムへの関与をさらに詳細に解明することが,薬物依存形成機構の解明と治療薬の開発に繋がると考えられる.
