Research Abstract |
ファイトプラズマ(Phytoplasma asteris)は、700種以上の植物に感染する植物病原細菌であり、農業生産上甚大な被害を引き起こす。ファイトプラズマは植物体内においては節部細胞に局在し、ヨコバイなどの吸汁性昆虫によって植物から植物へと伝搬される。このようにファイトプラズマは植物・昆虫という全く異なる宿主へと交互に感染する「ホストスイッチング」によって生活環を成り立たせており、その宿主適応メカニズムに興味が持たれている。本研究では、ファイトプラズマがどのように異なる2界の宿主環境を認識して遺伝子発現を制御しているのかを明らかにすることを目的とする。 昨年度は、本研究室で解読されたPhytoplasma asteris OY-Mの全ゲノム配列から、ファイトプラズマの転写因子としてRpoD及びFliAを特定し、これらの転写因子の宿主内における転写発現量を比較するため、RpoD及びFliAのリアルタイムPCR解析行った。その結果、RpoDの発現量は昆虫感染時に約4倍上昇していたことから、RpoDは昆虫宿主への適応に重要なOY遺伝子を制御していると考えられた。そこで本年度は、RpoDが昆虫感染時に発現量が上昇しているOY遺伝子(rrnB、rpsJ、gyrB)を制御しているかを調べるために、大腸菌内にRpoD発現ベクター及びこれらのOY遺伝子のプロモーター制御下にルシフェラーゼを組み込んだベクターを導入し、RpoDとプロモーターの結合能を解析した。その結果、RpoDを発現させた場合のみルシフェラーゼ活性が観察されたことから、RpoDはrrnB、rpsJ、gyrBのプロモーターを認識し、遺伝子発現を誘導することが明らかとなった。本研究より、RpoDは昆虫感染時に発現量が上昇しているOY遺伝子を正に制御していることが明らかとなり、RpoDはOYの宿主転換に伴う遺伝子発現制御に重要な役割を担うことが示唆された。
