Research Abstract

ボディープランの中での「シグナル分子班」のこの班での研究はかなり充実した成果内容となっている。浅島はアクチビンやレチノイン酸time-lay法によって膵臓形成をin vitroで行って、そこからインスリンやグルカゴンなどのホルモンを分泌する内分泌器官を未分化細胞から初めてつくった。またアクチビンによって活性化される新規の遺伝子としてMig30,XSIP-1などの初期発生に重要な遺伝子をクローニングし解析した。上野はBMPの標的遺伝子の解析を行って、Msx-1、VPMsx-1との関係を明らかにしたし、wntとTGF-βのクロストークについても証明した。また、線虫C.eleganceの体長を調節するリガンドCet-1の作用とその標的遺伝子としてlon-1をクローニングし、そのメカニズムを明らかにした。井出はニワトリやマウスの肢芽形成に関与する因子としてfgf-10とHoxa13,msx-1が大きな役割を担うことを明らかにした。野地はニワトリの胚が形成において特に四肢筋肉形成におけるcdf-8の役割を明らかにするためにトランスジェニックマウスを作製した。岡野は哺乳類神経幹細胞の自己複製に関してMsilとNotchシグナリングに注目して大きな成果をあげた。MsilはRNA結合性タンパク質であり、神経幹細胞に強く発現している。また、このMsi-1がNumbやHubと深く関与していることもわかった。佐藤はホヤを用いて脊索形成に関して、内胚葉分化とβカテニンとの関係や内胚葉からのシグナル分子としてのbFGFの同定、Brachyury遺伝子の標的遺伝子としてnetrin,prickle相同遺伝子、ERM、レプリカンなどそのカスケードを明らかにした。これらは更に新しくTbx-6の解析につながっていった。内山は、新しくTbx-6のクローニングと解析をした。
