Research Abstract |
The agent (prion) of transmissible spongiform encephalopathies (prion diseases) including scrape in sheep, bovine spongiform encephalopathy, and Creutzfeldt-Jakob disease (CJD) in human does not possess the agent-specific nucleic acid as a genome. Therefore genome typing and/or antigenic typing are not applicable for distinction of prion strains. However, prion strains can be distinguishable to some extent by biological and biochemical properties. In this study, we attempted to elucidate whether diversity of sheep prion strains exist in Japan. In addition, we analyzed genetic factor(s) other than PrP which will influence biological properties of prions. Eleven sheep scrapie occurred in Japan were examined in the transmissibility to mice and the relative protease-resistance of sheep PrPィイD2scィエD2, accumulatedin brains. s sheep prions exist in the field of Japan. Although characterization of the biological properties of sheep prions in the field has been reported and revealed diversity of sheep prions in UK, this is the first report which elucidated the diversity of sheep prions in Japan. The results obtained here will be useful to assess the possible risk for the spread of sheep prion in Japan to other species. A particular genotype of apolipoprotein E (ApoE) is known as a risk factor for Alzheimer's disease. It is also suggested that a particular genotype of ApoE might influence the disease type of CJD. Thus we examined whether there exists any relation between sheep ApoE and prion diversity or disease type. First we cloned genomic DNA and cDNA of sheep ApoE gene and determined the nucleotide sequence of them, and we identified three sheep ApoE genotypes.
