Research Abstract |
To investigate the applicability and practicality of Y-STRs, DNA was extracted from fo1lowing materials. 1) blood stains consisting of healthy unrelated Japanese, 2) vaginal swabs collected from 15 female cadavers subjected to a forensic autopsy, 3) the bone marrow obtained from the cadaver sunken under the sea for about one year, 4) the dental pulp of teeth incinerated in a dental furnace for 2min at various temerature. DNA was extracted with 150 mu1 Chelex Conventional Phenol-chloroform extraction method was applied to the bone marrow sample. PRC was carried out in 25 ゚C1 reaction voltmes, using 3 or 5ng of template DNA.The reaction condition were : 94゚C for 1min, 55゚C for 1min, 72゚C for lmin30sec for 30 cycles. Electrophoretic separation of PCR products was performed by high resolution polyacrylamide gel electrophoresis. Allele frequencies for DYS39O was as follows. 8 : 0.017 ; 9 : 0.153 ; 10 : 0.246 ; 11 : 0.288 ; 12 : 0.237 ; 13 : 0.051 ; 14 : 0.008. DYS389I : 8 : 0.018 ; 9 : 0.161 ; 10 : 0.268 ; 11 : 0.527 ; 12 : 0.027, DYS389II : 23 : 0.029 ; 24 : 0.105 ; 25 : 0.229 ; 26 : 0.324 ; 27 : 0.248 ; 28 : 0.048 ; 29 : 0.010 ; 30 : 0.010. Discrimination powers were calculated as 0.774 for DYS39O, 0.624 for DYS389I and 0.767 for DYS389II.Whenever a female cadaver subjected to a forensic autopsy, we take a vaginal swab and make some preparations with HE staining. The swabs were dry up and keep at room temperature. In 4 of 15 feamal autopsy cases, we could amplified DYS 390. There are good corresponding results between the amplification of DYS39O and the histological detection of sperm that is authentic evidence of exsistence of semen. We could amplified and typed the DYS39O using DNA cttained from the decomposed bone marrow of cadaver sunken under the sea for about 1 year. We also could amplified DYS 390 in all samples incinerated at up to 300゚C for 2min.
