Research Abstract

ヒト肝のエストロジェン代謝に焦点を絞って、ヒト由来の酵素を用いて代謝的活性化反応に関与するP450分子種を明らかにした。さらにP450酵素分子種毎の発現量に特徴のあるサンプルでの代謝産物と反応を触媒する主要なP450酵素分子種との関係に注目して個人差を整理して研究を進め、ヒト肝ミクロゾームと対比させることにより以下の成果を得た。第一に、エストロジェンとして内因性ホルモンからエストロン、エストラジオールを選び、ヒト肝ミクロゾームと反応させたのち、カテコールエストロジェントと16α水酸化体に着目して、代謝生成物を高速液体クロマトグラフィを用いて検出する系を整備した。第二に、ヒト肝ミクロゾームを用いてエストロン、エストラジオールの代謝物生成反応を調べ、代謝活性の個人差を検討した。他にも各P450酵素分子腫の指標となる典型的な薬物代謝活性を測定した。さらに免疫化学的に定量した各P450酸素蛋白の発現量とこれらの代謝活性との相関について検討を行った。ヒト肝ミクロゾームの活性に対する酸素阻害剤や特異抗体の影響を調べる実験や、ヒト遺伝子発現薬物代謝酵素を用いる実験、さらに遺伝子発現系を酵素源にした反応速度論的解析を行い、これらの内因性ホルモンの代謝的活性化に関与するCYP1A2とCYP3A4の役割を明らかにした。
