Research Abstract

脊椎動物の性成熟を支配する脳-脳下垂体-生殖腺軸(BPG-axis)の活動は, 春機発動の開始に伴い活性化すると考えられているが, 魚類ではその詳細は明らかでない。これまでBPG-axisの最上位における生殖制御因子として生殖腺刺激ホルモン放出ホルモン(GnRH)が知られていたが, 近年, 哺乳類において, GnRHの分泌を促して性成熟の引金を引く因子, キスペプチン(Kiss)が発見された。急速に進展している哺乳類でのKiss研究に比べ, 魚類のKiss研究は少なく, 現在, メダカ, ゼブラフィッシュ, キンギョ, フグやシーバスでの研究があるに過ぎず, その機能もほとんど明らかでない。本研究では, 春機発動機構解明のための解析ツールと全生活史にわたる飼育実験系が整備されているマサバを用いて, 期間内(平成23年9月~25年8月)に, KissによるGnRH制御機構, およびマサバへのKiss投与による春機発動促進効果を明かにする。
本年度(平成25年4月~平成25年9月)は, 2種のKiss受容体(KissR1, KissR2)の遺伝子クローニングを行い, 合成したマサバKiss1-15およびKiss2-12をリガンドとしたレポーター遺伝子アッセイを行った。その成果、KissR1はKiss1-15の、またKissR2はKiss2-12の固有な受容体であり、それぞれのKiss受容体へのシグナルは、PKC/MAPKs経路で伝達されることが明らかとなった。
