無脊椎動物の脳神経系と生殖腺におけるステロイドの生合成と生理作用に関する研究

Research Abstract

無脊椎動物の生殖腺においてステロイドが合成されていることは示唆されていたが、脳神経系や生殖腺でのステロイドの生合成経路やステロイド合成酵素に関する体系だった知見は極めて乏しい状況にあった。このような背景から本研究課題では無脊椎動物の脳神経系と生殖腺におけるステロイドの生合成経路を解析し、その合成酵素を同定することを目的とした。これまでの本研究課題による2年間の研究により、無脊椎動物のタコの脳神経系、ヒトデの神経系と生殖腺においてステロイドが生合成されていることを明らかにしている。本研究課題の最終年度である本年度は、無脊椎動物のステロイド合成酵素を同定することを目的とした。ヒトデの放射神経、卵巣、濾胞細胞などを用いて、次世代シークエンサーによるde novoシーケンシングを行った結果、脊椎動物と同様にシトクロムP450酵素系やhydroxysteroid dehydrogenase系のステロイド合成に関与する一連の酵素を同定することが出来た。また高速液体クロマトグラフ質量分析計(LC-MS)を用いて無脊椎動物の神経系で合成されるニューロステロイドの構造解析も実施した。これらの一連の解析により、ヒトデとタコの脳神経系や生殖腺におけるステロイド合成経路の大略を明らかにすることが出来た。
本研究により得られた知見は、無脊椎動物の内分泌学において学術的価値の非常に高いものである。今後、本研究課題の成果をもとに、無脊椎動物の神経内分泌学、生殖内分泌学の研究が大いに進展することが期待できる。
