Research Abstract

1)ptsG mRNA分解におけるdegradosome/enolaseの役割
代謝ストレスに応答したptsG mRNAの分解を担うRNase Eは細胞内で解糖系酵素であるenolaseなどと結合しRNA degradosomeを形成している。RNA degradosomeを形成できないRNase EのC末端領域欠損変異株の解析から、ptsG mRNAにはRNA degradosomeが必要であること、RNA degradosomeの構成因子であるPNPase(pnp), RhlB RNA helicase(rhlB),およびenolase(eno)の変異株の解析から、RNA degradosomeに組み込まれたenolaseがRNase EによるptsG mRNAの分解の制御に関わっていると結論した。これは、RNA degradosomeの構成成分であるenolaseの役割を明らかにした初めての例である。
2)ptsG mRNA分解におけるsmall RNA/Hfqの関与とmRNAの膜への局在の役割
代謝ストレスに応答したptsG mRNAの分解にはRNAシャペロンHfqが関与していることを見いだした。このことから特定のアンチセンスsmall RNAの関与が予想されたが、米国のグループが、機能不明であったsmall RNAの1つRyaA(SgrS)が、このRNAであることを明らかにした。これらのことから、代謝ストレスに応答したptsG mRNAの分解は、SgrS/HfqによるptsG mRNAへのターゲティングとそれに続くRNA degradosomeによる作用であることが明らかとなった。さらに、SgrS/HfqのptsG mRNAへのターゲティングにはptsG mRNAの膜へ局在化が必要であること、ptsG mRNAの膜へ局在化は翻訳の効率を低下させることによりSgrS/Hfqの作用を容易にする結果であることを明らかにした。
