Research Abstract

1.Thymidine phosphorylase(TP)とD-dRibは、HIF-1αのユビキチネーションを亢進して低酸素下でのHIF-1αのレベルを低下させる。HIF-1αは、アポトーシスをひき起こす因子であるBNIP3を誘導することが知られているが、低酸素下でBNIP3の発現が亢進し、TP、D-dRibがBNIP3の発現を抑制することがわかった。さらにD-dRibの立体異性体2-deoxy-L-ribose(L-dRib)はBNIP3の発現を亢進した。
2.TPcDNAをトランスフェクトしてTPを高発現したJurkat細胞は、ノコダゾール、ビンクリスチン、ビンブラスチン、パクリタキセルに耐性を示した。これらの抗癌剤はBcl-2をリン酸化し、FasLの発現レベルを抑制するが、TPはこのリン酸化を抑制し、FasLの抑制を阻止した。これらのTPの作用がマイクロチューブルに結合する抗癌剤に対してTP発現細胞を耐性することが考えられた。
3.L-dRibがTPを発現している腫瘍の浸潤をin vitroでもin vivoでも阻止すること、L-dRibがTP発現腫瘍で発現の亢進しているMMP-1とMMP-9の発現を抑制することを示した。
4.TPは癌組織に浸潤するマクロファージや樹状細胞にも高く発現していることから免疫応答にもなんらかの機能をもつことが予想された。TPノックアウトマウスでは、Th1免疫反応抑制作用のあるIL10の血中濃度が低下しており、癌細胞でもTP発現とIL-10の発現が関連していることが判明した。
