Research Abstract |
New photo-trigger methods were developed to study the protein dynamics and the energy changes during the chemical reactions of the proteins. 1. Protein dynamic of carboxymyoglobin was studied using the photo-dissociation reaction of the ligand. The dynamics and the energy change were revealed from nanosecond to second time range. 2. A pH jump method was developed to detect the protein structural change induced by the pH jump. The mechanism of the chemical reaction was studied. 3. Dynamics of apo-myogobin induced by the pH jump reaction was revealed. 4. Protein dynamics of cytochrome c was induced by photo-reduction method with a nanosecond pulsed laser. Thestructural change of cytochrome c after the photo-reduction was detected in terms of the volume change and the diffusion coefficient. 5. The photo-isomerization reaction of Photoactive Yellow Protein was used to initiate the cyclic reaction. A large temperature dependent volume change and change of the diffusion coefficient were discovered for the first time. The role of amino acids was revealed using the mutation technique. 6. A new reaction intermediate of octopus rhodopsin was discovered and the energy scheme of the reaction was determined.
