Research Abstract |
Auto-grafting of fat pads has a long history in plastic and reconstructive surgery for augmentation of lost soft tissue. Many uses of autogenous fat grafts, pearls and semi liquid adipose tissue in the treatment of depressed regions or scars in the facial area have been reported. Despite the enthusiasm for improved operation of free fat-autografts, however, researchers have been disappointed by progressive absorption of the graft with time. It has been accepted that the commitment of adipoblasts, unipotential precursor of adipocytes, completes during the embryonic development. Postnatal supply of adipoblasts has not been Expected. This is the reason why the surgery for argumentation of lost fat tissue has been focused only on transplantation of pre-formed fat pads. dy and that they can migrate, proliferate and maturate to form : fat pads with desired size and at desired site. Since this result implies a novel technique of augmenting lost j soft tissue, we here explore for the possibility of taking advantage of such adipocyte precursor cells by isolating and culturing them ex vivo as the autogenous cell for fat pad augmentation. In Yamaguchi et al, (2000), we have analyzed the structure and function of the G domain of laminin α4 chain which is synthesized by both adipocyte precursor cells and endothelial cells for the construction of the basemenf membrane structure between these tow types of cells. In Toriyama and Kawaguchi (2001), we reviewed our success of de novo adipogenesis in mice by injection of Matrigel. In Tajima et al, (2001), we found accelerated adipose conversion of 3T3-L1 fibroblasts under hypoxia due to stimulated type IV collagen synthesis. In Goto et al, (2001), we analyzed intracellular process of the assembly of Drosophila laminin α, β.γ Chains taking advantage of chain-specific RNA interference. In Kitagawa and Yamashita (2001), we introduced our recent data on the function of laminin a chain G domain in the peripheral connective tissue.
