Research Abstract |
A histological examination was conducted to assess the possibility of gonadal pathologies in four populations of konoshiro gizzard shad from Japan. Fish were collected off Haneda and off Futtsu in Tokyo Bay, off Zushi in Sagami Bay, and in Hamanako Sound. The location off Haneda corresponds to the site where feminized flounder have been previously reported. We also analyzed serum steroids in fish and muscle concentration of nonylphenol in fish. Intersex characteristics, such as the presence of perinucleolar oocytes, were found in 3 out of 73 males collected off Haneda but not in males from the other sites. Congestion-like pathologies were found in the ovaries of 2 females collected off Haneda and 2 from Sound Hamanako. Neither of these pathologies was correlated with the serum steroid hormone levels or with the size of the fish. The average concentration of nonylphenol in males collected off Haneda was significantly larger than that of males from Sagami Bay. Collectively, these findings suggest the possibility of endocrine disruption of normal gametogenesis in fish from some areas of Tokyo Bay and that this problem could be more widespread than currently recognized. An automated enantiomeric isolation system was developed using a gas chromatograph-preparative fraction collector to separate the enantiomers of chiral compounds. This system eliminate the need for a manual collection procedure after separation by gas chromatography. To evaluate the usefulness of this system we applied it to the separation and collection of the enantiomers of. -hexachlorocyclohexane, and we' measured the estrogenic potency of each enantiomer. Both enantiomers of. -HCH showed similar estrogenic activity. We concluded that this analytical method is suitable for the determination of the estrogenic activity of other chiral compounds. Such determinations are essential because enantiomers differ considerably in how they accumulate in organisms and decompose in the environment.
