Research Abstract |
Wistar male rat was inhaled toluene (1,500ppm) for 4 hours a day. We measured the weight of the rat, the testis and the epididymis. There were no significant difference in each weight in the short-term inhalation experiment. In the long-tern inhalation, body weight has decreased significantly, however, the testis and the epididymis were maintained. We examined the testis and the epididymis immunohistoohemically. HSP70 and c-Fos were not clearly observed in the control and the inhalation group. The cell damage was hot obviously. TGF-β1 immunohistochemically decreased in the testis and the epididymis of the inhalation group compared with the control. It was suspected to cause the disorder in the maturity process of the sperm. PCNA was observed by the spermatogonia and the primary spermatocytes of the testis, and the epididymis ducts. A significant difference of the PCNA-immunoreactivity was not admitted between the inhalation group and the control. It was suspected that the toluene has not caused the disorder to the division and the proliferation of the spermatogonia and the primary spermatocytes. Immunohistochemically, 8-OH-dG was increased in the spermatogonia in the testis. The amount of 8-OH-dG in the testis increased, but not significantly. The generation oxixidative DNA injury was suggested, and the influence on a reproductive function was thought. In the epididymis, the amount of 8-OH-dG has decreased. The 4-HNE was not observed in the control and the inhalation group in the testis. LPO of the testis showed quantitatively no significant difference between the control and the inhalation group. The lipid peroxidation in the testis was not clear. By toluene, the positive reaction of SOD has not increased obviously. That is, oxidative damage of DNA increased but it was thought that SOD had not been activated. It was suggested that the active oxygen elimination systems other than SOD had acted.
