Research Abstract |
1. By transplanting human melanoma cell clones transferred with expression vector carrying Has1,2 which codes hyaluronate synthetase, we obtained following results concerning in vivo phenotypes of the cells. (1) In addition to our previous report that Has1,2 transfected W793, a human melanoma cell line, was enhanced in cell motility(J.I.D , 1995), we confirmed the phenomenon by in dependent and different type of Has1,2 expression vector. (2) When we i.p. transplanted the newly established transfectants into immunodeficient nude mice (Balb/c), they formed tumor nodules 16-18 weeks after the transplantation with ascites. As for the mock transfectants, that was not the case even one year after the transplantation. By s.c. transplantation of the cells, we confirmed the tumor formation only in Has1, 2 transfectants. 2. Immunohistochemical staining of human melanoma tissues has been unsuccessful due to the lack of sufficiently specific antibodies of Has1, 2. 3. According to the results show in (1), resistance to apoptosis was regarded as one of the main of phenotypes acquired in the transfectants. When we investigated ASC gene (found by ourselves in 1999, JBC), it was very frequently and specifically down-regulated in human melanoma tissues.
