Localization of fibroblast specific protein 1 (FSP1) in kidney was studied by immunohistochemistry using anti-FSP1 polyclonal antibody. Renal biopsy specimens were obtained from 320 patients with various kinds of glomerulonephritis (IgA nephropathy, 142 patients ; membranous nephropathy, 28 patients; focal and segmental glomeruloscrelosis, 36 patients; minimal change nephritic syndrome, 31 patients ; lupus nephritis, 41 patients; crescentic glomerulonephritis 42 patients). FSP1 positive fibroblasts accumulated in the area showing severe interstitial fibrosis. Some tubular epithelial cells in the fibrosis area also expressed FSP1. The number of FSP1 positive cells was positively correlated with the area of fibrosis, which was measured using standard point-counting method (r = 0.84, p < 0.0001). The number of FSP1 positive cells was positively correlated with serum creatinine (r = 0.67, p < 0.0001) and negatively correlated with creatinine clearance (r = -0.71, p < 0.0001). We also examined glomerular FSP1 expression. In patients with crescentic glomerulonephritis, FSP1 was localized in visceral epithelial cells and in crescents. Levels of FSP1 expression in visceral epithelial cells in patients with focal and segmental glomeruloscrelosis was significantly higher than those in patients with minimal change nephritic syndrome.
