Research Abstract |
We have found the presence of oxidized protein hydrolase in the membranes in human erythrocytes. In this study, the following investigation was done. (1)The enzyme was isolated from human erythrocytes, characterized as a serine protease, and named oxidized protein hydrolase (OPH). (2)Cleaving sites of oxidized protein by OPH were clarified. (3)OPH was distributed in a wide variety of tissues and cells (4)COS-7 cells with highly expressing OPH(COS-7-OPH) were prepared, in which OPH was present uniformly in the cytosol. (5)COS-7-OPH was highly resistant to oxidative stress, indicating that OPH protected the cells from oxidative stress. (6)COS-7-OPH was resistant to oxidative stress in the presence of MG-132 that specifically inhibits proteasome. Involvement of Hsp70 in the activity of OPH was suggested.
