Research Abstract

腸炎ビブリオの分泌するthermostable direct hemolysin(TDH)、TDH-related hemolysin(TRH)は、腸管上皮細胞細胞内Ca2+濃度をprotein kinase C依存性に上昇させ、apical側に存在するCa2+依存性Cl-チャネルを活性化させることによりCl-分泌を亢進することを見出した。活性化されるCa2+依存性Cl-チャネルは細胞膜電位-60mVで21pSの外向き整流性を持っていた。またこのチャネルの活性化は、TDHやTRHによる直接作用ではなくレセプター等を介した間接的な作用であると考えられる。
さらにTDHやTRHの腸管上皮細胞に対する影響は、腸管上皮細胞の分化の程度によって感受性がことなり、より分化した腸管上皮細胞ほど感受性が高いことが分かった。このことは腸管ではbrush borderに近い上皮細胞ほど影響を受けやすいことが示唆された。
またTDHは上皮細胞内に取り込まれてアポトーシスを誘導することを明らかにした。このアポトーシスはエンドサイトーシスを阻害することにより抑制されることより、TDHはエンドサイトーシスにより細胞内に取り込まれることが示唆された。
