Research Abstract

DNaseI多型は対立遺伝子頻度のバラツキが少ない個人識別能の極めて高い遺伝マーカーである。予備実験によって、報告者らは唾液中にもDNaseI多型が検出できることを見出した。また本研究において、生唾液や唾液斑からのDNaseI型判定法を確立し、DNaseI型を唾液(唾液斑)からの有効な個人識別マーカーとして活用し得ることを証明した。
本年度は、上記の知見をさらに有用なものとするため、DNaseI多型に関する集団遺伝学的調査を拡大して行った。
日本の異なる9地域(旭川、青森、仙台、前橋、東京、福井、米子、松山および久留米地区)に在住している集団についてDNaseI多型を調査した。各地域在住者集団について判定したDNaseI型表現型の分布はすべてHardy-Weinberg平衡を示し、さらに各集団間には表現型分布に有意な相違は認められなかった。そこで、これら集団でのデータを合わせて、日本人集団(計2302人)におけるDNaseI型の6個の対立遺伝子頻度を求めると、DNASE1^*1、^*2、^*3、^*4、^*5および^*6の頻度はそれぞれ0.5467、0.4467、0.0074、0.0009、0.0002および0.0002と算定された。これとは対照的に、ドイツ人集団では異なる2地域(ミュンヘンおよびミュンスター地区)在住者集団間でDNaseI型表現型分布に有意な相違が認められた。従って、DNaseI多型の成立過程において人種間で異なる機構が作用したものと考えられた。次に、各々の地域在住者集団におけるDNaseI型を鳥瞰的に検討するとDNASE1^*2の頻度は青森(0.4750)および旭川(0.4747)地区在住者集団で最も高く、他方久留米地区在住者集団で最も低い値(0.4446)を示し、北から南に向けてDNASE1^*2頻度が減少する傾向が認められた。そこで各地域の緯度差とDNASE1^*2頻度との間の相関を統計学的に検討したところ、DNASE1^*2頻度に関して南北間に地理的勾配の存在することが判明した。
