Research Abstract

1 VIPによるヒト表皮細胞のcAMP産生促進とVIP receptor type I (VIP1R) antagonistによる,抑制について
VIPがVIP receptorを介して働くかどうかを検討するために有棘細胞癌株(DJM-1細胞)と正常表皮細胞にVIPを加えて,cAMPの産生について検討した.両細胞とも0.01μMから1μMのVIP刺激10分後に有意にcAMPの上昇がみられた.それは,VIP receptor 1R antagonistで抑制された.
2 VIP1R発現に及ぼすサイトカインの影響
DJM-1細胞の培養液にIL-4,TNF-α,IFN-γを加え,各時間後(0-48h)にVIP1RをNorthern blotで検討した.それぞれのサイトカインによってパターンは異なるがVIP1R mRNAの増加がみられた.
3 VIP刺激による表皮細胞からのサイトカイン、ケモカインの分泌とVIP1R antagonistによる抑制について
VIP(0.1nMから1μM)刺激により,DJM-1細胞におけるIL-1α,IL-6,IL-8,TNF-αとRANTESの産生についてELISAにより検討した.VIP刺激によりDJM-1からIL-6およびIL-8,RANTESの産生が有意に促進された.その反応はVIP1R antagonistによって抑制された.
今回の検討では,マスト細胞やリンパ球など真皮に存在する細胞から放出されるサイトカインが,表皮細胞のVIP receptorの発現の制御に関わっていると考えられた.また,DJM-1細胞にVIPを加えると,cAMPの産生が増強され,IL-6,IL-8,RANTESの産生が促進されたが,この反応はVIP1R antagonistによって抑制されたことから,VIP1Rを介して誘導されると考えられた.
