Research Abstract

本研究は不安障害の発症・病態に関与する候補遺伝子を同定し、機能解析を行うことを目的とした。我々はまず、新たな病因候補遺伝子として1)コレシストキニン系-CCK遺伝子およびCCKB受容体遺伝子、2)アデノシン(AD)系-A2a受容体遺伝子に注目した。1)(a)CCK遺伝子については、新たにshort tandem repeat(STR)と-345G>Cの2つの変異を同定した。さらに特定のallele lengthを持つ群およびSTR(379-383bp)-(-188G)-(-36T)haplotypeが不安障害患者に多く認められることを報告した。
1)(b)CCKB受容体遺伝子については、新たに3263G>C、3264A>Gの2つの変異を同定した。また、この遺伝子については約400bp上流に存在する繰り返し配列(CT)n多型と不安障害との間の関連が報告されていることから、これら既報の多型について日本人不安障害患者での検討を行ったが、関連研究では否定的であった。2)AD系ではA2a受容体について不安障害への関与が示唆されている1083C>Tを含む4箇所の遺伝子変異について関連研究を行い、日本人不安障害患者ではいずれの変異も疾患への関与の可能性は低いことを報告した。これらの結果から、CCK遺伝子の不安障害への関与が示唆された。
また、その他にもadenylate cyclase type 9(ADCY9)遺伝子、myo-inositol monophosphatase 2(IMPA2)遺伝子、tyrosine hydroxylase(TH)遺伝子、dual specificity MAP kinase phosphatase 6(DUSP6)遺伝子など、さまざまな精神疾患候補遺伝子について、不安障害をはじめとする精神疾患への関与の可能性を検討し、報告した。
