Research Abstract

(1)PP5/TFPI-2の臨床検体を用いた検討:産科疾患の胎盤(妊娠中毒症、常位胎盤早期剥離、子宮内胎児発育不全、血液疾患合併妊娠などの凝固異常が示唆される症例)についてPP5/TFPI-2の免疫染色やin situ hybridization法を用いてPP5/TFPI-2の生理機能について検討する目的で、現在インフォームドコンセントのもと、検体収集中の段階である。血小板減少性紫斑病の2検体についてはPP5/TFPI-2の免疫染色にて正常胎盤と差異を認めなかった。婦人科癌組織においても、PP5/TFPI-2の発現の様式と臨床病期・組織型・治療法(抗癌剤の効果の有無)・予後との関連についてなど解析中であるが、まだ症例数が少ないため、結論に至っていない。
(2)PP5/TFPI-2蛋白の細胞内の局在について:PP5/TFPI-2の生理機能を探る目的で、正常胎盤を用いてnano-gold法によりPP5/TFPI-2の免疫電顕を施行し、PP5/TFPI-2蛋白の細胞内の局在について検討したところ、PP5/TFPI-2蛋白はsyncytiotrophoblastのERと微絨毛の先端に認められた。またPP5/TFPI-2がヘパリンに結合する性格を利用して、初期胎盤(絨毛)をヘパリン添加溶液で培養したところ、ヘパリン濃度依存性に培養液中に検出されるようになった。これらから、PP5/TFPI-2蛋白はsyncytiotrophoblastの微絨毛の表面に存在し、先端から絨毛間腔へと絶えず産生・分泌され、絨毛間腔での物質交換や抗凝固作用を担っている可能性が考えられた。
