Research Abstract

損傷塩基除去反応の遷移状態を不安定化するヌクレオシドアナログとして、フラノース環2'β位にフッ素原子を導入した7-メチル-2'-デオキシグアノシン(1)を含むDNAを合成した。D-ribofuranose 1,3,5-tribenzoateを出発原料とし、(diethylamino)sulfur trifluoride(DAST)で処理し、2β位にフッ素原子を導入した。つづいて、HBr酢酸で処理して1-bromo誘導体へと導いた。このものをNaH存在下、2-amino-6-chloropurineと反応させβ-D-arabinofuranosylpurine誘導体を合成した。このものをsodium benzoxideで処理して6-benzyloxy-β-D-arabinofuranosylpurine誘導体へと導いた。アミノ基をphenoxyacetyl基で保護した後に、ベンジル基を除去した。続いて、CH_3Iで処理して、7-メチル誘導体とした。ジメトキシトリチル化、亜リン酸化を行ってアミダイトユニットを合成した。DNA自動合成機により5'-d(GCTTCGAGTAAXTTGGACACTATCC)-3'および3'-d(CGAAGCTCATTYAACCTGTGATAGG)-5'を合成した。ここでXは1、YはCあるいはTを示す。合成したDNAの構造は、DNAをヌクレアーゼおよびフォスファターゼでヌクレオシドへと加水分解し、このものと別途合成した1のC18逆相HPLCでの保持時間を比較することにより確認した。
