Research Abstract

Until recently, because of the shortage of the knowledge about biochemical and/or genetic characters, we have not been able to evaluate the usage of reductive TCA cycle for the production of useful materials. To solve this question, through characterizing the key enzymes of the cycle (Pyruvate : ferredoxin oxidoreductase, and 2-Oxoglutarate : ferredoxin oxidoreductase), basic biochemical information about the cycle for the production was collected and evaluation was carried out.
Actual results were as follows ;
(1) POR of Hydrogenobacter thermophilus, whose subunit composition was reported to be α・β・γ・δ,was actually composed of five subunits.
(2) Two sets of genes for OGOR were obtained and sequenced. Interestingly, the gene was located very closely with different direction of transcription. The genes were just separated by 〜100bp.
(3) The genes for OGOR seemed to be regulated by the aerobic/anaerobic condition of the cells.
(4) Experiments for RT-PCR for the evaluation of reductive TCA cycle in natural environments were set up. However, distinctive results have not been obtained.
