Research Abstract

申請者は,真正粘菌Physarum polycephalumで,ミトコンドリアプラスミド(mF)を見いだしている.本研究課題の目的は,(1)このmFプラスミドの起源とミトコンドリアへの侵入・伝播・成立の分子機構とその存在意義を明らかにするとともに,(2)mFプラスミドミの水平伝播能を利用し,ミトコンドリア用のベクター開発を目指すことにあった.mFは,mtDNAの一部(mID, 479bp)とほぼ完全に相同な領域(pID, 475bp)をもっており,この間の相同組換えによって,mtDNA内に組み込まれる.mIDがmtDNAに残されたmFの痕跡とすると(pID→mID), mFは,(i)P.polycephalumの種の確立する以前にミトコンドリアに侵入していたか,(ii)種の確立以後に極めて速やかに種全体に伝播したと考えられる.逆に, pIDがmtDNAに由来するものなら(mID→pID),(iii)mFは極最近になってP.polycephalumに侵入した可能性が高い.
収集の結果,P.polycephalumの由来の異なる粘菌アメーバを30株以上を保有することができた.本年度は,それぞれのmIDとpIDの塩基配列決定を引き続き行うとともに,近縁種の収集にも力を入れた.1)PCR法を用いて,組換え型,pIDとmIDの有無を調べた.また,mIDとpIDは全て塩基配列を決定し,塩基置換の種類と有無によってmIDとpIDの系譜図を作成している.これによって,pID→mIDとmID→pIDのいずれかが決定できるものと思われる.2)P.polycephalumおよびその近縁種を用いて,mFプラスミドの水平伝播の可能性を探るとともに,mFプラスミドを改変し,mFプラスミドの伝播に果たす末端タンパク質(TP)の役割に注目し,薬剤耐性マーカーの付加やpID改変によるベクター化の可能性を探っている.
