Research Abstract

リボゾームはタンパク合成の際に非常に重要な役割を果たしており、そのため、その異常は細胞にとって重大な影響を及ぼすものと考えられる。特に、発がん過程は細胞増殖に異常を来たした結果であり、そこにはリボゾームの関与も考えられる。しかしながら、これまでにこの点を明らかにした研究は行われていない。Fas刺激により、28SのリボゾームRNAが特異的に分解されるが、今回は、リボゾームタンパクの発現の面から検討した。これまでに我々が作成した21種類のヒトリボゾームタンパクを特異的に認識する抗体を用い、乳癌細胞においてFas刺激による影響を検討した結果、21種の抗体のうち、S11抗体がMCF-7細胞において特異的に発現低下を示していた。このS11の発現低下は、これまでの検討では、発現の変化は癌細胞間において不均一に観察されていた。今回観察されたこのS11の発現低下が転写レベルなのか、翻訳レベルなのか、或いはタンパクそのものの安定性や特異的分解によるのかは不明であり、また、発がん過程におけるこの発現低下の意味合いについても未解明である。今後、さらなる検討を加える予定である。
また、この他、14-3-3、p75NTR、EXTL3/EXTR1など、種々のアポトーシスに関連する分子についても検討を加えた。
本研究の過程において、リボゾームのサブユニットのユビキチンによる分解の過程を効率よく検出する電気泳動システムも開発したので、今後の研究に大いに役立つものと考えられる。
