Outline of Annual Research Achievements

本研究は，有害微細藻Heterocapsa circularisquamaによる水産業被害を軽減するべく，赤潮崩壊に関わるウイルスの影響、特にウイルス抵抗性を分子細胞学の視点で解析することを目的としている。一方、研究開始以降、ウイルス研究を完全無菌系で行う必要性、H. circularisquamaでの生理解析の難しさに直面し、基盤が整備されている珪藻について研究を進め、その情報をH. circularisquamaにフィードバックし成果を挙げてきた。本年度の計画では、ウイルス抵抗性細胞で特異的に発言する遺伝子があるかどうかを、珪藻の発現遺伝子から探索した。結果的にウイルス抵抗性細胞で発現の高い3つの遺伝子を同定し、これらが細胞膜近傍シグナル伝達分子をコードしていることを突き止めた。この解析により、ウイルス感染・抵抗性に細胞表面の細胞内シグナル伝達経路が関わる可能性が示唆され、ウイルス抵抗性関わる遺伝子の同定に大きな道筋をつけることができた。本年度はさらに宿主－ウイルス間の感染性の検証の為、ウイルス特異性に絞ったの解析を実施した。本年度までに117株の宿主珪藻と278株のウイルス株を分離しており、本解析ではこれらを対戦させ、感染成立の可否でパターン化した。その結果、RNAウイルスは感染性で2パターンに分かれるが、DNAウイルスは複数の感染パターンに分かれる事を突き止めた。これらのウイルスは同海域に生息していることから、同じ海域に生息する宿主・ウイルス個体群に多様性があり、これが海域でウイルスによって宿主個体群が死滅しない為の生存戦略「個体群レベルのウイルス抵抗性機構」として働いている事が示唆された。研究当初は「細胞性ウイルス抵抗機構」の解析を主に実施していたが、「個体群レベルでのウイルス抵抗機構」も重要である事を明確にすることになり、今後のウイルス研究の発展に貢献をした。
