Research Abstract

今年度、配列特異的アルキル化能を有したヘアピン型ピロール(Py)-イミダゾール(Im)ポリアミドの分子設計を確立し、Fmoc固相合成法と組み合わせることによって生物実験への供給経路を確立した。中でも、合成したアルキル化ポリアミドを用いて、標的配列特異的なアルキル化を行い、遺伝子機能発現に与える効果や細胞増殖阻害活性との相関性の評価を進めた。
その結果、標的とする塩基配列をタンパク質の遺伝情報が集約されているコーディング領域に設定し、その標的配列に対する特異的アルキル化によるタンパクの発現制御をヒト細胞実験によって確認した。また、ヒトがん細胞に対する遺伝子発現に対する効果を、ヘアピン型Py-Imポリアミドを用いて、DNAチップによって解析した結果、配列認識能の違いによる遺伝子発現の抑制や活性化に変化が現れることを確認した。加えて、39種類のヒトがん細胞スクリーニングパネル評価をすすめることによって、分子量が1しか違わない非常に構造の類似した2種の異なる配列特異性をもつアルキル化ポリアミド間に、配列特異性の差異に由来する細胞増殖阻害活性の差が観察された。
ラットを使ったポリアミド分子の細胞および核への透過性や臓器移行性の確認も進み、アルキル化ポリアミドを用いたゼノクラフトマウス実験による抗癌活性の評価も始めている。将来的には、薬剤としての最適化を進めながら、得られた生物活性情報を総合的に判断し、特定がんに対して特異的に作用するテーラーメード抗がん剤を実現する。
