Research Abstract

【目的】
我々はこれまで、抗癌剤耐性因子の一つであるGlutathione S-transefrase(GST)-πの活性を特異的に阻害するグルタチオンアナログγ-Glutamyl-S-(benzyl)cysteinyl phenylglycine hexadecylethyl diester(HGBP)を合成し、この化合物がin vitro及びin vivoにおいて種々の培養癌細胞の抗癌剤感受性を高めることを明らかにした。本研究では、HGBPの白血球増加作用を検討した。
【方法と結果】
(1)ラットにHGBP(10〜30mg/kg)を連日6日間投与し、経時的に採血して白血球数(分画)、赤血球数、血小板数を検討したところ、いずれの投与群においても好中球数がdose-dependentに増加した。同様にHGBPを連日投与し、骨髄におけるmyeloid cellsを検討したところ、阻害剤投与群では投与前に比べて有意なmyeloid cellsの増加を認めた。
(2)常法によりマウス骨髄を抽出し、IL-3及びSCFとともにGST-π阻害剤をそれぞれ10^<-9>〜10^<-5>M添加して軟寒天培地にて培養し、CFU-GM, CFU-E, CFU-Megなどを検討した。その結果、CFU-GMはdose-dependentに増加し、10^<-6>添加した群ではG-CSF 1ng/ml投与群と同様の効果が得られた。従って、GST-π阻害剤はG-CSFと同様に骨髄中のmyeloid lineageのprogenitar cellに直接作用して分化増殖を促進することが示唆された。
(3)5-FU投与骨髄抑制ラットを用いてHGBPの造血促進効果を検討したところ、HGBP投与群では好中球数がdose-dependentに増加するとともに、好中球がrecoverするまでの期間を短縮し、positive controlであるG-CSF投与群と同様の効果を有することが示された。
【結論】
GST-πに対する特異的阻害剤HGBPは、耐性を克服するのみならず、骨髄抑制をレスキューする効果もあることが示唆された。
