Research Abstract |
1.The mammalian endolymphatic sac(ES) is dramatically involved in regulation of endolymph homeostasis(endolymph composition and endolymph volume). The aim of this study is to investigate the involvement of ES epithelial cells in endolymph homeostasis. To examine the early events of endocytosis of ES epithelial cells, we injected tracers of endocytosis(MPO,HRP,CF) into the guinea pig ES in situ and examined using electron microscopy. There are ribosome/rough endoplasmic reticulum rich cells(RRC) and mitochondria rich cells(MRC) in guinea pig ES epithelium. RRC endocytosed plenty of HRP with the uncoated vesicle, and medium amount of CF with the coated vesicle but a little MPO. Several RRC did not show any endocytic activity. 2.Examination of the morphology of ES epithelium of rat. There are RRC,MRC and melanocytes. RRC are main epithelial cells and have well-developed rER, and Golgi apparatus and many round〜oval electron-dense granule(melanosomes). At the apical surface, there is a few short microvilli. MRC has a numerous oval〜elongated mitochondria in the whole cytoplasm and few melanosomes. At the apical surface, there are numerous long, slender microvilli. Apical part of cell is protruding into the ES lumen. Cell have bottle-necked figure. Melanocytes are occasionally observed at the base of epithelium. The long processes of cells are extended among MRC and RRC. It looks like that melanocytes distribute the melanin granules to MRC and RRC. 3.Development of ES. To examine the morphological change of ES during, the development, we made the 20μm serial sections of rat ES from newborn to 1 year old and made three dimensional model of ES. Most astonishing observation was that ES of newborn and infant were equal, but that of aged rat were flattened sac. It was supposed that ES function were active during development and gradually become inactive according aging.
