Research Abstract |
THE PURPOSE OF THIS STUDY WAS TO EXAMINE (1) MORE SENSITIVE AND RELIABLE ANALYTICAL METHODS OF INTERNAL EXPOSURE MARKERS AND DNA ADDUCTS, (2) INDIVIDUAL FACTORS AFFECTING ON LEVELS OF INTERNAL EXPOSURE MARKERS AND DNA ADDUCTS, (3) VALIDITY OF THESE BIOMARKERS IN OCCUPATIONAL SETTINGS, COKE OVEN WORKERS EXPOSED TO POLYCYCLIC AROMATIC HYDROCARBONS AT A HIGH LEVEL. DEVELOPMENT OF ANALYTICAL METHODS OF INTERNAL EXPOSURE MARKERS AND DNA ADDUCTS IS ONGOING. WE MADE A PROTOCOL OF THE VALIDATION STUDY OF THESE BIOMARKERS, OBTAINED IRB APPROVAL, STARTED TO OBTAIN BIOLOGICAL SAMPLES IN COKE OVEN WORKERS. CONCERNING INDIVIDUAL VARIATION OF THESE MARKERS, WE OBSERVED THAT 1-HYDROXYPYRENEANDAROMATIC DNAADDUCTS WEREAFFECTED BY CYP1A1 OR NAT2 GENOTYPES. 8-HYDROXYDEOXYGUANOSINE IN WHITE BLOOD CELL-DNA WAS NOT ELEVATED IN SUBJECTS EXPOSED TO POLYCYCLIC AROMATIC HYDROCARBONS ATA HIGH LEVEL. THE MESSENGER RNA OF CYP1B1 IN WHITE BLOOD CELL WAS OBSERVED TO RELATE WITH SMOKING IN COKE OVEN WORKERS.
