Research Abstract |
The differences of components of glycolipids between cord and adult blood were studied. After extraction of glycolipids from plasma and red blood cell, the components were analyzed by TLC, TLC-immunostaining and HPLC technique. 1. In TLC analysis, glycolipids were visualized with orcinol reagent. Three bands were mainly detected in plasma and five-six bands in red blood cell having no relation with ABO blood type and Lewis blood type. But the other staining bands of low Rf values were detected in adult plasma but not in cord plasma. 2. In immunostaining, biotin-anti-Le^a, biotin-anti-Le^b and biotin-anti-Urex europeus UEA1 were used. Owing to cross reaction among these antibodies, antibody specific bands did not observed. 3. HPLC analysis was carried out using 4-aminobenzoic acid octyl ester labelled kit for relation contents of oligosaccharides, and using 4-aminobenzoic acid ethylester labelled kit for sugar compositions. The number of mail oligomer was almost same (oligomer 1 and 2) in both cord and adult blood, but oligomers in cord blood were more large. The contents of sugar composition were determined as mainly Gal and Glc in cord and adult plasma, in and cord and adult red blood cell were mainly Gal, Glc and GlcNAc. The amount of Glc was more higher than Gal in adult plasma. In both cord and adult red blood cell, the amount of Glc and Gal was same level.
