Research Abstract |
In this study, we aimed to find a new gene which may fuse to API2 gene in gastric MALT lymphoma. In the first year of the project, we examined mRNA in human biopsy specimens from 5 MALT lymphomas by using primers designed for API2 gene and putative new fused gene. However, we failed to find specific bands, partly because the primers were not specific enough to detect the putative gene. In the second year, to get a proper primer with high specificity and the best experimental condition, we designed many primers and continued series of trials and errors on mRNA from a gastric cancer cell line-AGS cells and normal human biopsy and surgical specimens of the stomach in preliminary studies. As a result, we could not detect a new gene that may play a role in the pathogenesis of MALT lymphoma within the project period. We intend to purify specific cDNA by nested PCR on PCR products and try to find an objective gene on the human MALT lymphoma tissue in the next year.
