Research Abstract |
Using reconstruction of the urinary bladder mucosa in three-dimensional collagen gel culture, we investigated the role of angiogenic factor in bladder cancer by a series of histological observation. Normal transitional epithelial cells isolated from porcine bladder were cultured on reconstructed lamia propia using fibroblasts.in type I collagen gel. The inoculated transitional epithelial cells grew, in a stratified fashion and closely resembled urothelium in viva Human urinary bladder carcinoma cell (HUBCC) line (HT-1197, RT-4, T24) was seeded onto this model. HT 1197 cells were infiltrated in lamia propia. Next, vascular endothelium and epidermoid growth factor (EGF) were added into the, collagen gel and cultured. But the proliferations of HUBCC were equal to the fibroblasts alone model. Then in order to evaluate the angiogenesis in vivo, we transplanted the reconstructed transitional epithelium HUBCC was seeded to rats and tried to observe the angiogenesis in histologically. However all rats died few days later from transplantation and the grafts did not adhere in autopsy. In vitro," HUBCC was seeded onto the reconstructed mucosa again and sections were analyzed for expression of vascular endothelial growth factor (VEGF) protein. However no VEGF protein expression was observed. And expression of hypoxia-inducible factor l a and thymidine phospholylase were not observed in Western blotting. We were not able to prove the angiogenesis in ladder cancer in this study, so we have to develop new technique.
