Research Abstract

We revealed that the neuron-restrictive silencer (NRS), which is one of the target genes of the neuron-restrictive silencer factor (NRSF), was located in the 3' untranslated region of the atrial natriuretic peptide (ANP) gene and medizted repression of ANP promotor activity in ventricular mayocytes (Kuwahara. K, MCB, 2001). Thus, it suggested that NRSF regulated not only the neuronal cells
Therefore, we established NRSF or NRSF-VP16, which has transcriptional activation domain of the virus, constitutive expression cell lines and used performed subtractive PCR by using two cell lines that the expression of endogenous target genes were decreasing or increasing to search new target genes. Consequently, the kinesin light chain (KIF2), which is concerned with the vesicle transport, Non-POU domain containing octermer-binding protein, which is the transcription factor, and so on, were obtained
On the other hands, as the result of DNA date base searching of human and mouse, we succeeded to obtain a lot of target genes over 200. The gene whichwas named as the candidate NRSF target gene, the gene which was concerned with monoamine oxicase A or B, which plays important role on neural functions, the molecules which have new ankyrin repeat proteins and Zn-finger transcription factor as the candidate NRSF target gene were included among them
These results shows that utilization of the target genes assessing system by using cell lines enables us to know actual NRSF target genes
