Research Abstract

(1)Keap1タンパク質の構造/機能連関の解析
Keap1の機能を解析するために,一過性共発現系を用いたレポーターアッセイ法により解析した.親電子性物質に反応するアミノ酸残基としてシステイン残基が考えられるので,システインをアラニンに変換した変異体を作製して親電子性物質に対する反応性に関して解析したところ、BTBドメインとDG.ドメインの間に存在するIVR(Intervening Region)中のシステイン残基に変異を導入した時に、親電子性物質に対する反応性は低下した.以上の結果から、親電子性物質のセンサーとしてKeap1のシステイン残基が考えられた.
(2)Keap1と相互作用しないNrf2タンパク質点変異体を用いた解析
線維芽細胞に安定遺伝子導入したNef2-EGFPは,プロテアソーム依存性に素早く分解される.Neh2領域中の蛋白質分解に必要なアミノ酸残基を同定するために,Neh2 N末端とNeh2 C末端とをそれぞれEGFPに融合した蛋白質を作製し,その蛋白質半減期を解析した.結果,Nrf2 N末端の領域をEGFPに融合した蛋白質は野生型と同様の半減期を示したが,Neh2 C末端をEGFPに融合した蛋白質は著明な安定化を示した.この領域は、ユビキチン化に必要とされるリジン残基を持っていないことから、Neh2によるプロテオソーム依存性の蛋白質分解は、ユビキチン非依存性の機構によることが考えられた.
