Research Abstract

本研究の目的は、パネート細胞に特異的な内因性抗菌ペプチドであるパネート細胞alpha-defensinのsubfamilyを生化学的方法で分離同定し、その機能を明らかにすることである。異所性に出現するパネート細胞のうち、胃の腸上皮化生粘膜のパネート細胞と潰瘍性大腸炎の大腸粘膜のパネート細胞に注目し、インフォームド・コンセントの下に材料を得て、それらに含まれるdefensinの性状と機能を検討した。その結果、胃粘膜の腸上皮化生でみられる胃パネート細胞からのわれわれが確立した方法を用いてdefensin分画を回収し、HD5を同定した。さらに、潰瘍性大腸炎の大腸粘膜にみられる大腸パネート細胞からdefensinを同定するために、パネート細胞の出現が確認された潰瘍性大腸炎の活動性炎症大腸粘膜または緩解期大腸粘膜から分離大腸陰窩を得た。大腸パネート細胞にalpha-defensin分画を同定した。これらのdefensin familyは小腸パネート細胞のそれと同様に、サルモネラ菌に対する殺菌活性を有していた。本研究で得られた各種蛋白検体をHPLC、電気泳動法、western blot法等により生化学的、免疫生化学的に解析して、それらの殺菌活性を同定、分離した。
本研究により、ヒトの消化管における自然免疫機構は、単に受動的なものではなく、defensinをはじめとする内因性抗菌ペプチドをエフェクターとする、すばやく強力で、よく制御された系であることが示唆された。
