Outline of Annual Research Achievements

水中で金属錯体を安定化させるには多座配位の活用がエントロピー的に有利であるものの、金属中心への電子供与はLewis酸性の低下にも繋がりかねない。そこで平面的な多座配位によって中心金属のd軌道、特にdz2軌道を制御する事が水存在下での不斉反応の鍵となると考え、xy方向での混成に加えdz2軌道と配位子自身のs, pz軌道との混成、またLewis塩基の配位を通じたz軸方向のエネルギー準位安定化を期待した触媒系の開発に引き続き邁進した。Jahn-Teller効果によって不安定化されているdz2軌道を活用すべく、銅(II)を基調とする触媒系による不斉ホウ素共役付加反応を拡張した。従来報告されてきた銅(I)触媒では厳密な無水条件及び強塩基の添加が必須、基質適用範囲は使用する不斉配位子の構造に強く依存していたのに対し、開発した触媒系は強塩基が不要で、驚くべき触媒活性を示しTOFの最高値は43200 h-1を記録した。C-エノラートを経るとされる銅(I)に対して銅(II)ではO-エノラート中間体が想定される為、これまでになく幅広い基質に対して高選択性を与える事にも繋がった。更に水中では加水分解を容易に起こしてしまう不飽和イミンに対してもその特徴を遺憾なく発揮し、ほぼ完璧な立体選択性(>99% ee)が得られた。また、酵素の構造的巧緻さを模倣した人工設計による新規キラル触媒の開発を目指した。構造設計に基づく特徴は、①水中で機能、②Lewis酸及びBronsted酸の三次元的配置による基質の同時活性化、③疎水性部位導入による効率的不斉反応場の形成、④Bronsted塩基の適所配置に基づくエノラートの効率的発生及びプロトンの効率的運搬である。活性官能基の三次元的な適所配置の結果、水中に分散する分子量800程度の人工設計分子に協同的触媒活性を付与する事に成功した。
