Research Abstract |
Despite recent development of a number of combined therapeutic approaches, the prognosis for pancreatic cancer is still extremely poor. Although, there are options of chemotherapies of Gemzar, 5-FU or other agents, an effective protocol has not been established yet. 5-FU is popular, however, the effective rate is 15-20%. Consequently, new treatment system including diagnosis is necessary to be developed. We developed a new diagnostic system using mutant replication-selective adenovirus encoding the green fluorcent protein (GFP). The purpose of this study is to assess the effect of develop the new gene diagnostic system by the utilizing videoscope. Furthermore, we used a selectively replication-competent adenovirus that can multiply only in abnormal p53 gene cells to induce the higher expression of GFP genes. This adenovirus can infect also the adjacent non-infected cancer cells, overcoming limitation of the transfer efficacy by a replication-incompetent virus. Three pancreatic cancer cell lines infected with the adenovirus encoding the GFP gene changed its fluorecence. These results strongly suggested that GFP gene diagnostic system is effective for intra-operative diagnosis of pancreatic cancer and using mutant adenovirus make this strategy much stronger.
