Research Abstract |
Chronological change in PPARγ expression was evaluated immunohistochemically. PPARγ was expressed in the cytosol of some epithelial cells, many of intraepithelial and lamina proprial lymphocytes. PPARγ expression decreased following ischemia/reperfusion injury. Pre-medication with PPARγ ligands prevented the decrease in PPARγ expression level. Interestingly, PPARγ was detected in the nuclei in the presence of PPARγ ligands. Administration of PPARγ ligand without I/R injury caused no change in PPARγ expression, while PPARγ ligand plus I/R injury brought about slight decrease in PPARγ expression. PPARγ expression in colonic biopsy specimens of ischemic colitis patients was examined immunohistochemically. As a whole, no remarkable change in PPARγ expression was noted in those samples. As a control, colonic tissues from ulcerative colitis (UC) and Crohn's disease (CD) patients were examined for PPARγ expression immunohistochemically. PPARγ expression was reduced in UC samples, whereas no significant change was noted in CD samples. PPARγ ligand pioglitazone was given to 8 patients with acute ischemic colitis, and the disease course was observed. Abdominal pain was relieved in all of the 8 patients after pioglitazone administration. PPARγ immunostaining of colonic biopsy samples in those patients revealed that PPARγ was expressed not only in the cytoplasm, but also in the nucleus of the epithelial cells.
