Research Abstract |
Hemoglobin gene was transferred to mouse liver to estimate the protective effect of newly expressed hemoglobin in the liver from ischemia-reperfusion injury. The adenovirus vector containing myoglobin gene was administered through the penial vein of C57BL mice and the liver samples were analyzed by RT-PCR and immunohistochemistry at 3 days after gene transfer. RT-PCR revealed the expression of mRNA of the myoglobin gene in the liver. Immunohistochemistry showed positive staining of myoglobin protein in the hepatocytes. The amount of ATP in the myoglobin-gene transferred liver was larger than those of the non infected group and the LacZ-introduced group. The hepatoduodenal ligaments of the gene-transferred and control mice were clamped for 30 minutes for total hepatic ischemia and the liver samples were collected at 24 hours after reperfusion. TUNEL staining revealed positive staining in the control mice while the gene-transferred mice showed a few TUNEL positive cells. Serum ALT increased in the control mice but it remained normal in the gene-transferred mice. However, Immunohistochemistry of bcl-2,Bel-XL and Bax showed no difference among these 3 groups. Gene transfer of hemoglobin was effective in protecting the liver form apoptosis after ischemia-reperfusion injury.
