Research Abstract |
We have made pnumoperitoneum model adjusting the intra-abdominal pressure of 8 mmHg in a rat showing a stable hemodynamics. We showed that solid organs (liver, kidney and spleen) blood flow reduced significantly during pneumoperitonium in contrast to gastrointestinal tract organs such as stomach and intesitine showing no significant reduction in blood flows proven by microsphere methods. The blood levels of antidiuretic hormone (ADH) significantly increased immediately after pneumoperitoneum. We conducted polymerase chain reaction (PCR) methods to investigate the effects of pneumoperitoneum on the body because this method showed the early expression of mRNA of various mediators such as TNF-alpha, IL-1 beta and IL6 which increased in the patients with trauma before any detection of these mediators in the bloodstream. Also we planed to study the effect of local anesthetics on the expression of mRNA of these mediators. However, we could not detect any expression of mRNA of these mediators not only in control model but also in pneumoperitoneum model including a model using local anesthetics. Several reasons were considered. This model might not be so invasive to prove the expression of these mediators in the level of mRNA. We concluded that this study did not show any expression of mRNA using PCR methods during pnuemoperitoneum.
