Research Abstract |
We have established T cell clones(TCC) derived from the aqueous humors(AH) and cerebrospinal fluids(CSF) of Vogt-Koyanagi-Harada(VKH) disease patients. The TCCs were established with limiting dilution method after transforming the T cells with Herpesvirus Saimiri(HVS) C488. Four hundred forty four TCCs derived from AH of 3 patients and 958 TCCs from CSF of 7 patients were obtained. Among those TCCs, randomly selected 21 TCCS derived from the AH and 9 TCCs from the CSF of patient 1were challenged with tyrosinase and/or TRP1 peptide mixture. Ten out of 16 CD4 positive TCCs from the AH and 2 out of 6 CD4 positive TCCs from the CSF were reactive against tyrosinase or TRP1. The cytokine production profile showed that all except one of the CD4^+, CD8 TCCs reactive to tyrosinase or TRP were Th1 type. It was strongly suggested that the T cells reactive against tyrosinase and/or TRP1 play an important role to the induction and formation of the inflammatory lesions in VKH disease.
